چکیده
Abstract CD8+ T cells and NK are the primary line of defense against tumor cells. However, microenvironment impairs their anti-tumor capabilities. One key functions impaired is ability to infiltrate tumor, essential allow direct contact with a target cell its killing. Kv1.3 KCa3.1 K +channels have multitude that been established in cells: regulates cytotoxicity chemotaxis. less known about Herein, we performed electrophysiological 3D chemotaxis experiments study activity functional role human were isolated from healthy donors (HD) head neck cancer (HNC) patients activated IL-2 IL-15 for 24–48 hr. Activation led an increase currents 49.6% at 24 hr 434.1% 48 . increased by 42.8% 54.2% Chemotaxis experiment showed move randomly collagen matrix migrated towards chemokine gradient presence CXCL10. Chemotaxis, but not random migration, was inhibited selective blocker TRAM-34 HD HNC patients. Shk-Dap22, blocker, did alter either or migration. Overall, reduced distance achieved 157% (4 donors) 89% (1 donor). These data highlight critical channels migration open possibility targeting facilitate infiltration

Phosphoinositide 3-kinase (PI3K), PTEN and localized phosphatidylinositol (3,4,5)-trisphosphate [PtdIns(3,4,5)P3] play key roles in chemotaxis, regulating cell motility by controlling the actin cytoskeleton in Dictyostelium and mammalian cells. PtdIns(3,4,5)P3, produced by PI3K, acts via diverse downstream signaling components, including the GTPase Rac, Arf-GTPases and the kinase Akt (PKB). It ...

Natural Killer (NK) cells are innate lymphocytes with an important role in the early defense against intracellular pathogens and against tumors. Like other immune cells, almost every aspects of their biology are regulated by cytokines. Interleukin (IL)-15 is pivotal for their development, homeostasis, and activation. Moreover, numerous other activating or inhibitory cytokines such as IL-2, IL-4...
متن کاملPost-transcriptional Regulation of NK Cell Activation
Natural killer (NK) cells play key roles in innate and adaptive immune defenses. NK cell responses are mediated by two major mechanisms: the direct cytolysis of target cells, and immune regulation by production of various cytokines. Many previous reports show that the complex NK cell activation process requires de novo gene expression regulated at both transcriptional and post-transcriptional l...
